In vivo or ex-vivo.2 In vivo methods involve injecting a bioengineered antigen, drug or other compound directly into the body, with the expectation that DC will respond to the engineered molecules more readily than they have to the natural tumor antigens. Some in vivo therapies aim to deliver the DC-activating antigen in novel form e.g., as part of a modified tumor cell or as "naked" DNA ; or vehicle e.g., fat emulsions or virus vectors ; . Other in vivo methods, assuming that angiogenesis factors or some other DC-suppressing agents are at work, are designed to circumvent those factors or reverse their inhibitory effects. We here present data on 17 patients with male LH-independent precocious puberty who have been analyzed for the presence of mutations of the LH receptor gene. We have developed a new assay for measuring epidermal growth factor receptor EGFR ; activation using the BRET ; technology, which directly measures the recruitment of signaling proteins to activated EGFR. Our results demonstrate that EGFR BRET assays precisely measure the pharmacology and signaling properties of EGFR expressed in HEK293T cells. EGFR BRET assays are highly sensitive to known EGFR ligands pEC50 EGF 10.1 + - 0.09 ; , consistent with previous pharmacological methods for measuring EGFR activation. We applied EGFR BRET assays to study the characteristics of somatic EGFR mutations that were recently identified in lung cancer. In agreement with recent reports, we detected constitutively active mutant EGFR isoforms, which predominantly signal through the PI3K Akt pathway. The EGFR inhibitors Iressa or Tarceva are several folds more potent in inhibiting constitutive activity of mutant EGFR isoforms compared to wild type EGFR. Notable, our results reveal that most of the mutant EGFR isoforms tested were significantly impaired in their response to EGF. The highest level of constitutive activity and nearly complete loss of EGF responsiveness was detected in isoforms, which carry the activating mutation L858R and the secondary resistance mutation T790M. In summary, our study reveals that somatic mutation in EGFR quantitatively differ in pharmacology and signaling properties, which suggest the possibility of differential clinical responsiveness to treatment with EGFR inhibitors. Furthermore, we demonstrate that the EGFR BRET assays are a useful tool to study the pharmacology of ligand-induced interaction between EGFR and signaling pathway specifying adapter proteins.

Called `a poor response' to gonadotrophins are still controversial. Identification of these patients is usually based on results of a previous IVF stimulation and the presence of one of the following characteristics: three or fewer recruited dominant follicles or collected oocytes; serum oestradiol concentrations lower than 300 or 500 pg ml at the time of HCG administration; In fact, these criteria are not universally accepted, and some authors include patients whose cycle was cancelled because of a spontaneous LH surge or who require a large dose of gonadotrophins. Is there any common feature in poor responders to gonadotrophins? In some of them, an incipient ovarian failure can be predicted by hormonal markers in basal and stimulated conditions; indeed, ovarian reserve and prediction of ovarian stimulation response have been, so far, evaluated by a single basal FSH determination during the early follicular phase Muasher et al., 1988; Scott et al., 1989, 1990; Hansen et al., 1996; Magarelli et al., 1996; Kim et al., 1997 ; or by dynamic challenge tests: clomiphene citrate Scott et al., 1995; Scott and Hofmann, 1995 ; or GnRH agonist stimulation test Galtier-Dereure et al., 1996; Yamashita et al., 1996 ; . Moreover, the prognostic value of day 3 plasma oestradiol determination has been reported Licciardi et al., 1995; Smotrich et al., 1995 ; . The large heterogeneity within the group of poor responders easily explains our current difficulties in managing ovarian stimulation in this group, and emphasizes that every new therapeutic approach must be analysed carefully, with special attention given to the criteria of patient inclusion. Keeping in mind these restrictions, let us examine the recent reports on adaptations of GnRHa protocols in this situation. Wenlock Edge, May 22nd 2002. A smallish group met at Harton Hollow to explore the surrounding woodlands in the light rain of a pleasant spring evening. Although this tiny Shropshire Wildlife Trust reserve is very well known, the extensive woods to the north and south are hardly recorded at all. We took the opportunity to go northwards to Harton Wood, but not without stopping on the edge of the reserve to photograph the Hybrid Avens, Geum x intermedium. There have only ever been eleven records of this plant in the county, so it is not common. The footpath through Harton Wood runs along the top of the slope between deciduous plantation in calcareous W8 ; woodland below, and conifer plantation in acid W10 ; woodland above. Unsurprisingly, the track itself and the occasional clearing were the best botanical locations. Wild Service Tree, Sorbus torminalis, was spotted by Henry Hand, giving us a new tetrad for the species. A small group of shrubs and suckers straddling the path was the only patch seen. John Clayfield found a clump of Carex pallescens, Pale Sedge, which was last recorded here by William Phillips in 1904. It turned out to be quite frequent along a recently felled area of forestry. To our disappointment, Paris quadrifolia, Herb Paris, eluded us but on our return to the car park we found that Audrey Ashwell had stopped by to leave a frond of Beech Fern, Phegopteris connectilis, as proof of its continuing existence at the Hazeldine Coppice. In total eighty-nine species were recorded for Harton Wood, which is a significant increase over the previous four.
