We are a technology company and everything we do is built upon the creative insights and technologies necessary to discover, formulate, devise manufacturing processes, manufacture and improve our medicines. Remodulin, a prostacyclin analog United Therapeutics has focused primarily on developing Remodulin as our lead product for treating pulmonary arterial hypertension, a life-threatening vascular disease that affects the blood vessels between the heart and lungs. Pulmonary arterial hypertension is characterized by the degradation of the blood vessel wall lining, the aggregation of platelets and the disruption of smooth muscle cell function. These conditions cause blockages and affect the ability of the blood vessels to dilate and then constrict as blood flows to the lungs. The resulting elevated pulmonary blood pressure causes increasing strain on the right side of the heart as it tries to pump blood to the lungs. It is estimated that there are between 50, 000 and 100, 000 individuals with pulmonary arterial hypertension in the United States alone. Pulmonary arterial hypertension is associated with reduced production of the natural hormone prostacyclin in the pulmonary blood vessels. As an analog of natural prostacyclin, Remodulin appears to dilate blood vessels where necessary, prevent platelet aggregation, and prevent proliferation of smooth muscle cells surrounding the vessels. In 13 patients for whom information about APOE allelic status was known, there was a correlation between the number of APOE4 alleles and the rate of change of the CDR sum-of-boxes scores r 0.59; P .03 ; but not the rate of change in ADAS-Cog total scores, MiniMental State Examination total scores, or Neuropsychiatric Inventory total scores. Adjusting for the number of APOE4 alleles in these patients eliminated all significant correlations between neuroanatomical measures and the rates of change in clinical outcome variables. Patterns of hippocampal surface variation associated with various rates of change of ADAS-Cog total scores are illustrated in the Figure. Patients with the most positive rates of change ie, worsening symptoms of dementia ; showed inward variation of the surface of the hippocampus, especially within the LZ and IMZ, relative to the entire population of patients. Conversely, patients with the most negative rates of change ie, improving symptoms of dementia ; showed an expanded hippocampal surface within the LZ and IMZ relative to the entire population of subjects!
Surgery commonly creates significant post-operative pain, although a few orthopedic procedures do not routinely warrant a `pre-emptive' nerve block. This concept was elucidated in an analysis of 1, 200 consecutive knee surgery out-patients.20 In this study, 543 patients underwent `less invasive' out-patient knee surgery, while the remainder underwent `more invasive' knee surgery. Forty-three per cent of the `less invasive' patients received nerve block s ; , although the use of nerve blocks in these patients was not associated with a reduction in symptoms, nursing interventions, or unplanned hospital admissions.20 Consequently, for `less invasive' knee surgery out-patients, nerve blocks should be reserved for situations of significant refractory postoperative pain, elevated pre-operative pain, 21 or intolerance to traditional oral analgesic techniques.

Testicular microlithiasis TM ; is a rare human pathologic condition characterized by calcifications within the seminiferous tubules, originating from degenerative intratubular cellular debris that becomes surrounded by concentric layers of stratified collagen fibers. TM has been associated with a wide variety of testicular pathologies, including primary testicular neoplasms and intratubular germ cell neoplasia ITGCN ; . Whether these associations are causal or coincidental is not known. We describe herein a case of TM in canine testicle whose controlateral cryptorchid testis developed a early seminoma associated with a Carcinoma in situ CIS ; -like pattern. A Siberian Husky dog 5 yrs old, 27 bw ; presented a 7 days history of abdominal pain. Since 18 month old the dog revealed abdominal cryptorchidism of the right testicle. Alpha-fetoprotein 31.33 ng ml ; and testosterone 7.7 ng ml ; were detected by EIA. A TM was diagnosed by high-frequency 5 7, 5 MHz ; testicular ultrasound that demonstrated multiple hyperechoic foci in the left testicle. After radical orchiectomy, the presence of TM was confirmed by routine histopathology. Several degree degenerative changes occurring in germ cells were associate to TM. In the controlateral atrophic undescendent testis, an early seminoma TS ; was detected, associated with an ITGCN showing a characteristic CISlike pattern. To our knowledge, the association of TM, TS and CIS has never been reported in dogs so far. The present paper further supports the opinion that dogs could be a very interesting animal model for human testicular disease and renagel.

From August 21, 2001, through February 1, 2002, 22 patients with a total of 28 target lesions were treated with oral rapamycin. Baseline characteristics are listed in Table 1. Patients had an average of 3.5 1.6 episodes of previous restenoses. Previous radiation failure occurred in 20 90.9% ; patients, and 2 9.1% ; patients were not candidates for radiation therapy both because of very small and tortuous target vessels not considered accessible by radiation delivery catheters and reserpine.
Transgenerational phenotype was transmitted through the male germ line 15 ; Fig. 1 ; . The phenotype of increased spermatogenic cell apoptosis and decreased sperm numbers and sperm motility was observed in greater than 90% of all males of all the generations examined. When the animals were allowed to age up to 1 yr, additional diseases developed including cancer, prostate disease, kidney disease, and immune cell defects Anway, M. D., and M. K. Skinner, submitted for publication ; . A high frequency of transmission was observed in all generations examined for all the disease states. The frequency of the transgenerational phenotype was such that a DNA sequence mutational event could not be involved. The random nature of a DNA sequence mutation has a phenotype typically less than 1%, and this often declines in subsequent generations 50, 104 ; . An epigenetic mechanism is involved because of the frequency of the phenotype. To support these conclusions, two genes were identified in the sperm that had altered methylation patterns associated with the transgenerational phenotype discussed 15 ; . Therefore, the endocrine disruptors appear to induce an epigenetic transgenerational disease condition for four generations through the male germ line 15 ; Fig. 1 ; . The epigenetics appears to involve altered DNA methylation. Although most genes get reset in early embryonic development, a subset of genes called imprinted genes maintains their DNA methylation pattern that appears to be permanently programmed. In contrast to all somatic cells, the primordial germ cells undergo a demethylation during migration and early colonization of the embryonic gonad, followed by a remethylation starting at the time of sex determination in a sex-specific manner 23, 24, 105 ; . The exposure of the pregnant mother at the time of sex determination appears to have altered the remethylation of the germ line and permanently reprogrammed the imprinted pattern of DNA methylation 15 ; . This provides a unique epigenetic mechanism to promote a transgenerational phenotype induced by an environmental factor.

The electric field very close to a fluid contact line located at the interface of two materials with differing dielectric constants is investigated analytically. This is accomplished via a conformal mapping of the solution domain, as well as series expansions of the electric potential very close to the contact line. The field within each material near the tri-phase boundary is examined and used to discuss the relative importance of such phenomena as charge trapping, corona discharge, local dielectric breakdown and finite fluid resistivity to contact angle saturation. 1: 33PM OC.00007 Numeric Simulation of EWOD and DEP Force Distributions , PATRICK YOUNG, ERIC BAIRD, KAMRAN MOHSENI, University of Colorado at Boulder -- Two primary methods for electrostatically actuating microdroplets in channels currently exist: Dielectrophoresis DEP ; for electrically insulating liquids, and electrowetting on dielectric EWOD ; for conducting fluids. In each case, a transverse electric field is used to create an electrostatic pressure for transporting individual liquid slugs. This paper examines the nature of the force distribution for both EWODand DEP-actuated droplets. The effects of system parameters such as dielectric constant, dielectric thickness, contact angle and channel height on the shape of the force density and its net intergral are considered. A comparison of the scaling properties of the DEP and EWOD methods for applications in digital microfluidics is presented and restasis.

Signs and symptoms of overdose with Remodulin during clinical trials are extensions of its dose-limiting pharmacologic effects and include flushing, headache, hypotension, nausea, vomiting, and diarrhea. Most events were selflimiting and resolved with reduction or withholding of Remodulin. In controlled clinical trials, seven patients received some level of overdose and in open-label follow-on treatment seven additional patients received an overdose; these occurrences resulted from accidental bolus administration of Remodulin, errors in pump programmed rate of administration, and prescription of an incorrect dose. In only two cases did excess delivery of Remodulin produce an event of substantial hemodynamic concern hypotension, near-syncope.
Treatment with 2 mg kg day, alternate-day, topical, replacement, or aerosolized steroid preparations is not a contraindication to varicella-containing vaccine. 2 Persons whose immunosuppressive therapy with steroids has been discontinued for 1 month 3 months for chemotherapy ; may be vaccinated and revlimid.
