Five patients died of treatment-related complications: neutropeniaassociated sepsis in three, pneumonia in one and electrolyte imbalance in one. Non-myeloid toxicities including nausea, vomiting, mucositis, alopecia, skin rashes, chemical conjunctivitis, central nervous system toxicity and elevation of serum transaminase and creatinine levels are shown in Table 2. Severe mucositis grade 3 or 4 ; was found in six patients. Mild to moderate skin rash was found in five patients. Elevation of the serum transaminase level was noted in six patients, including one grade 3 and one grade 4. Elevation of the serum creatinine level was noted in one patient, grade 2. Alopecia was found in all patients, grade 3 being seen in the majority. Infection was found in six patients, two with grade 3 and three with grade 4. Only one patient had transient chemical conjunctivitis, grade 2. No significant cardiac toxicity was found.

Results Effect of developmental stage on outcome of thawing A total of 4006 embryos was thawed in 1657 cycles. Among the thawed embryos, 562 were zygotes and 3444 were cleaved embryos. The large majority 91% ; of cleavage-stage embryos were frozen on day 2 n 3133 ; , and the remainder n 311 ; on day 3. The effect of the developmental stage of the zygotes and cleaved embryos on survival after thawing is summarized in Table I. The seminal plasma of several animal species contains a high molecular weight antifertility factor "Decapacitation Factor" ; . Recently, the presence of a similar compound in human seminal plasma was firmly established Reddy et al, 1979, J Reprod Fert, in press ; . The objective of the present study was to purify this factor from human seminal plasma and study some of its physiologic properties. The mouse in vitro fertilization system was used as an assay. A high molecular weight pellet was obtained from human seminal plasma by ultracentrifugation at 104, 000 g for 18 hours. The compound was further sequentially purified by CMCellulose, Concanavalin A, and Sepharose 6B chromatography. A 129-fold purification was achieved. The purified preparation showed a single band on polyacrylamide gel electrophoresis. Amino acid analysis showed the presence of a relatively high number of valine, leucine, and isoleucine residues. At a concentration of 45 g ml, the compound caused 5O% inhibition of in vitro fertilization when added to capacitated mouse spermatozoa. The inhibition of fertilization was dose dependent and the effect could be reversed by incubating the treated spermatozoa in capacitation medium. Further biochemical and physiologic properties of the compound, including its mechanism of action, are being investigated. Supported by NIH grant HO 09868.

Materials and Methods Eligibility Eligible patients were between 15 and 59 years of age with newly diagnosed AML, morphologically defined as FAB M0 to M7 the French-American-British classification of acute leukemias 8 ; . Long-term arsenic exposure is associated with the development of skin cancer, while arsenic trioxide has also been effective in treating acute promyelocytic leukemia APL ; . The present study was to investigate the molecular mechanisms underlying the diverse arsenic toxicity. At low dose 0.1-1 microM ; , arsenic induced the proliferation and wound closure of human keratinocyte HaCaT. This effect was further potentiated by the growth factors, TGF and TGF, but not by the cytokine TNF. We suggest that arsenic signal cooperates with the TGF and TGF signals to accelerate wound closure and that arsenic may accomplish its effects through the activation of the TNF signaling pathways, therefore, it does not further potentiate TNF stimulated wound closure. This suggestion was supported by the findings that evodiamine, a chemical inhibitor of the TNF effector NF-B, completely abolished arsenic-induced keratinocyte wound closure. At high concentrations 5-20 microM ; , arsenic inhibited cell proliferation and wound closure and it caused the induction of hemeoxygenase-1 expression and lactate dehydrogenase LDH ; release. To investigate the role of TNF-induced IB kinases IKK ; and the NF-B pathway in high dose arsenic toxicity, we examined cells deficient in the IKKs. Compared to the wild type cells, the IKK- and IKK-null cells were more susceptible to arsenic induced apoptotic cell death. Arsenic induced HO-1 expression in the IKK-null cells to the same degree as in the wild type cells; however, it caused a much stronger and prolonged induction of the c-Jun N-terminal kinases and damiana.
