On Monday, December 11th, Newark's math league teams hosted the 2nd competition of the year for Region 1: Caravel, Glasgow, Middletown, Red Lion Christian Academy, St. Andrew's Academy, St. George's VoTech and Newark. It is tradition to supply snacks and drinks before the competition. Due to obvious circumstances, the math league had to look elsewhere for help. Many people stepped up. I would like to thank a few. Kelly Roarty, mother of Jessica on Senior High Team C, supplied all the sodas and waters for approximately 150 people. Ms Tami Lunsford and the Newark Key Club provided so many cookies, pretzels and chips that there were leftovers. Laura Bedford, mother of Jaclyn on 9th Grade Team B, shopped and provided over 200 cookies. Tiffany Wen, a senior on Team A, baked brownies. Leo Marianiello, a senior on Team A, organized the food so that everyone was served. He began the process and then we had to evacuate the building for awhile. When we were allowed back in, Leo continued the process flawlessly. Newark's Math League teams and Mr. Koliss would like to say "Thank you" to all who made it.

The intent and object of the adoption of the Voluntary Plan is to help only those applicants whose conduct, both past and present, indicates that they were or are denied insurance for reasons other than those attributed to absence of proper appreciation of their responsibilities to the State, and to their fellow men. If it were the intent to interpret 'good faith' as meaning only the absence of enumerated offenses, it would not be possible to deny the application of the plan to the automobile owner or operator who, although not guilty of enumerated offenses, is engaged in a business definitely illegal and contrary to the expressed policy of the State. Certainly the plan is not intended to keep on the highways of the State persons whose use of the automobile is in the business of smuggling, illicit sale Of merchandise, or promoting illegal gambling. Neither is it the intent of the plan to help the applicant who misrepresents the facts in order to mislead insurers and those charged with the administration of the plan. The plan should be construed and administered as in the nature of equitable relief and the ordinary principles of equitable relief should apply. The plan is not intended to aid the carrying on of illicit trades and practices and neither is it for the benefits of the persons who, by misrepresentation or perjury, conceal material facts. Instances of false statements that have arisen in the administration of the plan involve apparently deliberate omission of statements pertaining to prior convictions or suspension of licenses, false statements respecting ownership of motor vehicles, false statements respecting the registration of vehicles and the license of operators, and false statements respecting the occurrence of prior accidents. In many such instances the omission of such essential information required in the application form, or actual misrepresentation, apparently indicated that the risks were eligible for coverage under the plan. However, investigation by the designated carrier and reconciliation of developed facts with the records available from the Motor Vehicle Bureau of the State disclosed apparently deliberate attempts on the part of the applicant to obtain coverage through false statements. In these and all similar instances it is the opinion of the Committee that such applicants are not exercising that.

Objective : Liver biopsy for the assessment of liver fibrosis in HCV patients is an invasive, unpleasant and suboptimal method. Therefore the aim of this study was to discover and identify new potential biomarkers for liver fibrosis in HCV patients using proteomic technology. Methods : A HCV patient cohort n 30 ; was selected and matched according to age at biopsy ; , weight, alcohol consumption andduration of infection. The cohort was divided according to the 5 fibrosis stages Metavir F0 : none n 6 F1 mild n 6 F2 moderate n 6 F3 advanced n 6 F4 cirrhosis n 6 ; . Serum samples were depleted from albumin and IgG and were used for theanalysis.The proteins were separated using two-dimensional polyacrylamide gel electrophoresis 2D-PAGE ; pH-gradient 4-7, 10% SDS-PAGE ; and visualized with a fluorescent protein marker, Sypro Ruby Molecular Probes ; . Analysis of the gels was performed using PD-Quest Bio-Rad ; . The different gels were matched and regrouped to the 5 fibrosis stages F0-F4 ; and inflammatory activities A0-A3 ; . The groups were analyzed with the Student's t-test and the Mann-Whitney U-test p 0, 05 ; . Proteins were identified using two complementary mass spectrometry MS ; techniques, namely MALDI-Q-TOF MS and ESI-Q-TOF MS MS on-line coupled to a nano-Liquid Chromatography LC ; system. Results : After gel analysis a total of 1356 separated spots have been matched between all gels. 117 protein spots were differently expressed of which 91 spots between different fibrosis stages F0-F4 ; and 26 spots between inflammatory activities A0-A4 ; . Among these spots alpha-2-macroglobulin F0 F4 ; , serum albumin F0 F1 ; F0 haptoglobin F1 F4 ; were identified, which are currently used in non-invasive alternatives. Conclusions : During this discovery phase of our proteomics analysis, protein patterns have been identified that can discriminate different stages of liver fibrosis. Among the 117 differently expressed proteins, so-called surrogate markers of non-invasive alternatives have been identified. This indicates that our approach has potential to discover and identify biomarkers for different stages of liver fibrosis and posses possibly new unknown marker proteins. However the identified proteins need to be further investigated to characterize their involvement in the pathology and their potential as true fibrosis markers.

Bleeding is very unusual in young babies. Characteristically children with severe haemophilia have their first bleed when they first start to crawl or walk from around nine months onwards. Sixteen out of 85 Sprague-Dawley rats 18.8 per cent ; developed solid medullary tumors in the left ocular cavity within 74 to 288 days after a single intravitreous inoculation of 0.015 to 0.03 ml. of fluid containing human adenovirus type 12, 103-5 to 10i5 TCIDso HeLa cells per 0.1 ml. The left eye was inoculated within 24 hours after birth. Tumor-bearing rats were perfused with Karnovsky's fixative and their enucleated eyes were subjected to epon-embedded thin sectioning. The remarkably uniform microscopic appearance of all cases resembled human retinoblastoma with incomplete rosettes. Ultrastructurally, the tumor cells forming incomplete rosettes and perivascular wreaths possessed oval or slightly indented nuclei without noticeable nuclear membrane abnormalities. A slender luminal pole of cytoplasm in each cell contained poorly differentiated organelles. One cilium per cell with a 9 + pattern of doublets associated with a pair of centrioles was detectable in all cases. A strong tendency for many tumor cells to undergo bizarre ganglioneuronic maturation was also characteristic. The role of the CaR in regulating PTH has been demonstrated in in vitro studies with calcimimetic agents 41 ; . In early study, Hammerland et al. 46 ; expressed bovine and human CaRs in Xenopus laevis oocytes. They showed that increasing the extracellular concentration of calcium led to an increase in the chloride ion current across the cell membrane. The early type II calcimimetics, NPS R-467 and NPS R-568, potentiated the effects of extracellular calcium and other type I agents. Neither calcimimetic had any effect on the chloride current in the absence of extracellular calcium. Further studies from the same group showed that NPS R-467 and NPS R-568 increased the sensitivity of bovine parathyroid cells to extracellular calcium 85 ; . R-enantiomers of both compounds in nanomolar concentrations inhibited PTH release and shifted the calcium-PTH response curve to the left. In vivo experiments performed in rats showed that when NPS R-568 was given by gavage to normal rats, there was a rapid decrease in serum PTH 36 ; . Minimum PTH levels were reached within 15 min of dosing and were followed by a decrease in serum calcium, which was expected after a marked reduction in PTH.

Conventional hyperimmune antisera raised against M. hyopneumoniae gave high background values in the ciliary binding assay and were not useful in this analysis data not shown ; . DNA sequence analysis of pISM2121 revealed an open reading frame coding for a 124.9-kDa protein. The open reading frame was defined based on the availability of an AUG start codon and upstream sequences that may function as transcriptional and translational start sequences. Analysis of the P97 DNA sequence revealed several interesting features. First, the protein had remarkably few hydrophobic regions, with a single 17-amino-acid hydrophobic region sufficiently large to span a membrane in the amino terminus. The major antigenic regions were predicted to be near the C terminus of the protein, although there were putative antigenic regions scattered throughout the N-terminal portion of the molecule as well data not shown ; . In support of this hypothesis, the Tn1000 insertional mutagenesis data indicates that the MAb binding epitope lies between the Tn1000 insertions at positions 16.13 and 1.14. It is possible that this region of the gene is duplicated in the chromosome to give rise to the multiple bands observed in immunoblot profiles of M. hyopneumoniae proteins with MAb F1B6 35 ; . Data from our hybridization using three internal probes of the P97 gene sequence suggested the possibility of a single duplication of P97 Fig. 5 ; , but this would not completely account for the complex immunoblot pattern with MAb F1B6 Fig. 2 ; , and thus, this issue remains unresolved. A 5-amino-acid repeating motif of 15 repeats with a consensus sequence of AAKPV E was identified in the region of the translated sequence containing amino acids 814 to 887 Fig. 3, nucleotide positions 2440 to 2665 ; . A second repeating motif, of 10 amino acids, with four repeats of consensus sequence GTPNQGKKAE, was identified near the carboxy terminus at amino acids 981 to 1020 nucleotide positions 2941 to 3061 ; . Other than Mhp1 13 ; GenBank accession number U27294 ; , this repeat region showed no similarities to known proteins of other mycoplasma species, including Mycoplasma genitalium, for which the entire DNA sequence is known 7 ; . The insertion 16.13, located at position 3, 012 of the P97 gene sequence within the second repeat region of the carboxy terminus, resulted in a truncated gene product in E. coli of approximately 98 kDa, as compared to the full-length 124.9-kDa protein Fig. 2 ; . This proved unequivocally that pISM2121 coded for the gene product recognized by MAb F1B6 and that the protein was not an E. coli gene product induced by the presence of the recombinant plasmid. The gene product of insertion 16.13 also bound to swine cilia Fig. 4 ; , suggesting that the region downstream of this insertion may not participate in cilium binding. To rule out this possibility, all non-P97 downstream sequences were deleted to form plasmid pISM1228 and the recombinant products were tested for MAb reactivity and swine cilium binding activity. E. coli lysates containing pISM1228 had immunoblot profiles similar to those of E. coli lysates containing pISM2121 and pISM2159 Fig. 2 ; . Cilium binding activities were also similar data not shown ; . Alternate codon usage by mycoplasmas results in premature truncation of cloned gene products in E. coli. This unusual gene codon usage has the potential to prevent identification of specific genes by immunological approaches, particularly when MAbs are used to screen genomic libraries. In the studies presented here, MAb F1B6 failed to identify recombinant and codeine.
